GC/MS ANALYSIS AND ANTIMICROBIAL ACTIVITY OF PENTACYCLIC TRITERPENOIDS ISOLATED FROM COMBRETUM GLUTINOSUM PERR. EX. DC. STEM BARK
Abstract
Purification of combined fractions 23-30obtained  from column chromatography of methanol extract of Combretum glutinosum Perr.ex.Dc stem bark was carried out using preparative thin layer chromatography with ethylacetate as developing solvent. This led to the isolation of four components C-1, C-2, C-3and C-4 (RF=0.817, 0.412, 0.147 and 0.088 respectively, three(C-1, C-2 and C-4) of which afforded a better yield and were further investigated using IR, GC/MS and were tested for antimicrobial activity. A pentacyclic triterpenoid of the type friedelan-3-one was identified as the major constituent hereby reported in C. glutinosum for the first time. Other compounds identified were stigmast-4-en-3-one, lupan-3-one and lupeol. The later though present in a lesser quantity manifested a greater antimicrobial potency. The isolated components (C-1,C-2 and C-4) exhibited significant antimicrobial activity against Klebsiellae pneumoniae with MIC of 1.625x103  µg/ml, 3.0x103 µg/ml, 3.25x103 µg/ml respectively; and were resisted by Corynebacterium ulcerans, Escherichia coli and Neisseria gonorrhea. The antimicrobial property observed may be attributed to the identified triterpenoids in the isolated components. Further spectroscopic analyses using 1Hnmr and 13Cnmr is however required to confirm the structure of these active principles.  Keywords: Combretum glutinosum Perr.ex.
